Properties

Applications

Our Abpromise guarantee covers the use of ab45420 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
ICC/IF Use a concentration of 10 µg/ml.
IHC-P Use a concentration of 5 µg/ml.
WB Use a concentration of 1 - 4 µg/ml. Detects a band of approximately 24 kDa (predicted molecular weight: 21 kDa).

Target

  • FunctionAssociates with IL12B to form the IL-23 interleukin, an heterodimeric cytokine which functions in innate and adaptive immunity. IL-23 may constitute with IL-17 an acute response to infection in peripheral tissues. IL-23 binds to an heterodimeric receptor complex composed of IL12RB1 and IL23R, activates the Jak-Stat signaling cascade, stimulates memory rather than naive T-cells and promotes production of proinflammatory cytokines. IL-23 induces autoimmune inflammation and thus may be responsible for autoimmune inflammatory diseases and may be important for tumorigenesis.
  • Tissue specificitySecreted by activated dendritic and phagocytic cells and keratinocytes. Also expressed by dermal Langerhans cells (at protein level).
  • Sequence similaritiesBelongs to the IL-6 superfamily.
  • Developmental stageExpressed by newborns dendritic cells.
  • Cellular localizationSecreted. Secreted upon association with IL12B.

  • Immunohistochemistry of IL-23 in human pancreas tissue with ab45420 at 5 µg/mL.

  • ab45420 at 10 µg/ml staining IL23 in Raji cells by ICC/IF
  • ab45420 staining IL23 P19 (green) in Human THP-1 cells by Immunocytochemistry/ Immunofluorescence.

Cells were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100 and blocked with 5% serum for 1 hour at 20°C. Samples were incubated with primary antibody (1/200 in 5% goat serum + 1 BSA) for 1 hour at 20°C. An AlexaFluor®488-conjugated goat anti-rabbit polyclonal IgG (1/500) was used as the secondary antibody.
