  • FunctionAssociates with IL12B to form the IL-23 interleukin, an heterodimeric cytokine which functions in innate and adaptive immunity. IL-23 may constitute with IL-17 an acute response to infection in peripheral tissues. IL-23 binds to an heterodimeric receptor complex composed of IL12RB1 and IL23R, activates the Jak-Stat signaling cascade, stimulates memory rather than naive T-cells and promotes production of proinflammatory cytokines. IL-23 induces autoimmune inflammation and thus may be responsible for autoimmune inflammatory diseases and may be important for tumorigenesis.

Anti-IL23 antibody [B-Z23] images

    Human monocytes were fixed in methanol, permeabilized using 0.1% Saponin/ PBS, blocked with 4% serum for 30 minutes at 25°C and then incubated with ab84471 at 5µg/ml for 48 hours at 37°C.

    Staining: IL-17 (red) GFP (green).

    We have examined the effect of IL-23 and in combination with anti-IL-23 on IL-17 production in monocytes. IL-23 leads to increased IL-17 secretion (Figure B and C) and this effect was abolished with anti-IL-23 (Figure D). IL-2 is part of our culture medium for monocytes. IL-2 stimulation serves as a basis to keep the cell alive. Thus almost every cytokine that we used is used in combination with IL-2. Because it is part of the medium, IL-2 can be omitted when specifying the stimulation. The cells were stained with the GFP (Green Fluorescent Protein), just to make them visible. We opted for GFP, because the cells are less affected by GFP as by dyes.The red color represents the IL-17 which was the target in this experiment. The secretion of IL-17 was tested with and without IL-23 and in combination with anti-IL-23. If the green of the GFP interferes, all images can be shown without the green channel (red only).
