1. Antibody purification kits

To remove unwanted proteins or preservatives - up to 0.5 mg of antibody in 100-500 µl

Commercially available antibodies often contain substances (e.g. BSA, glycine, tris, azide) that interfere with your experiments.

Abcam's antibody purification kits quickly remove these contaminants. They can also be used to purify antibodies from crude samples such as ascites fluid or serum. The optimal volume of the sample to be purified is 100-500 µl and up to 500 µg of antibody can be purified in each run.

The method involves capture of the antibody on protein A, protein G or mouse specific resin and the removal of unwanted substances by a simple wash procedure, which is carried out in a standard microfuge. The purified product is then eluted and neutralized. This method cannot be used with samples containing relatively dilute antibody in large volumes (e.g. tissue culture supernatant).

Protein G resin has a high affinity for the Fc regions of IgG molecules from a variety of species (e.g. sheep, rat or goat). Protein A resin has a high affinity for the Fc regions of rabbit IgG molecules. For purifying mouse antibodies from crude samples such as ascites the Mouse Antibody purification kit is recommended with a high affinity for mouse IgG molecules (independent of the sub-isotype) and negligible binding to Bovine IgG. The Mouse Antibody purification kit is based on a proprietary affinity resin and is not suitable for use with antibodies from other species.

To purify tissue culture supernatant (TCS) - up to 5 mg of antibody in 10-50 ml

Abcam's antibody TCS purification kits are prepared by coupling highly purified protein A to agarose beads and can therefore be used to purify IgG fractions from supernatants. The antibody is captured on the TCS resin and unwanted substances are removed by a simple wash procedure. The purified product is then eluted and neutralized. Antibody TCS purification kits have been designed for tissue culture supernant, and can be used to purify up to 50 ml.

For the selective purification of mouse IgG from hybridoma supernatants without co-purifying bovine IgG, the Mouse TCS purification kit is recommended as protein G or protein A resin cannot selectively purify mouse IgG. The Mouse TCS purification kit has a higher affinity for mouse IgG molecules

To purify serum or ascites fluid - up to 20 mg of antibody

Abcam's antibody serum purification kits are prepared by coupling highly purified protein A or G to agarose beads and can therefore be used to purify IgG fractions from both serum and ascites fluid. The antibody is captured on the agarose beads and unwanted substances are removed by a simple wash procedure. The purified product is then eluted and neutralized. Antibody serum purification kits can purify up to 20 mg of antibody. The volume of sample required will depend on the host species and will be mentioned on each datasheet.

2. Antibody concentration kits

Antibodies are sometimes only available at low concentrations and often contain low molecular weight substances that interfere with your experiments.

Our antibody concentration kit allows for the quick and easy concentration of antibodies and proteins. Additionally, the kits can also be used to reduce the concentration of many unwanted additives often found in antibody solution such as azide, glycine or tris.

Abcam's antibody concentration kits use a simple spin column to easily and quickly remove excess buffer from the antibody thereby providing a more concentrated antibody solution. The kits also allow for a simple buffer exchange.
