Methylation of cytosine residues located on CpG islands in promoter regions is generally associated with transcription repression. Hydroxymethylation (hydroxylation and methylation) of cytosine residues in promoter regions is a recently discovered modification in animal cells which seems to play a role in DNA demethylation, although its function is still unclear. Bisulfite modification is used to determine the methylation pattern of DNA: DNA is treated with sulfites, a process which will convert cytosine residues to uracil while leaving 5-methylcytosine residues unaffected. Therefore, the bisulfite treatment will introduce specific changes in the DNA sequence depending on the methylation status of individual cytosine residues, producing single-nucleotide resolution information about the methylation status of the DNA sequence of interest.
