Feedback Form

All antibodies work in different combinations of applications, species and samples, so we encourage our customers to write a short Abreview, so that future customers can make an informed decision about the usefulness of a particular antibody for their specific experiments.

Application:

Immunofluorescence

Blooms Syndrome Protein Blm antibody (ab476)

Additional Data

Notes

The antibody worked beautifully for immunofluorescence using paraformaldehyde/Triton-X-100 fixation and an antibody dilution of 1:600 overnight at 4 degrees.

Dilution notes

Immunofluorescence 1:600

Application:

Immunofluorescence

Blooms Syndrome Protein Blm antibody (ab476)

Additional Data

Notes

The BLM antibody worked nicely in immunofluorescence. In western blot the BLM protein was also well visible although there was quite a lot of background.

Dilution notes

1:500 for immunofluorescence
1:2000 for Western blot

Abcam response

Thank you for your very useful feedback information. We would suggest testing out different blocking agents, and/or decreasing the concentration of the primary antibody.

Application:

Western blot

Blooms Syndrome Protein Blm antibody (ab476)

Additional Data

Notes

The Blm antibody worked well for western blotting, where
a number of background bands were also visible, with a whole cell lysate. However immunoprecipitation followed by
western blotting revealed a single band.

Notes

I used the Rb polyclonal to BLM (Bloom's syndrome protein) for immunofluorescence(1:500). This protocol worked great with a methanol fixation/permeablization, but not with a paraformaldehyde fixation and a triton X-100 permeablization. This antibody also worked well for my westerns (1:1000), although there was an extremely high amount of background bands. I wouldn't have been able to determine the BLM band without a negative control.

Application:

Western blot

Blooms Syndrome Protein Blm antibody (ab476)

Additional Data

Notes

ab476 is fine for doing western blot and immunostaining on culture cells. It was not successful to use ab476 to do coimmunoprecipitation.

Notes

This antibody works fine as well on mouse embryos by whole-mount immunohistochemistry.
At 1:100 dilution, still you can get nice staining.
