This antibody would not work for me in a simple two-step fluorescent protocol. Appears rather sensitive to overfixation. It works well, however, with tyramide signal amplification or standard DAB techniques. Here, I've used the Perkin Elmer TSA-Cy3 kit. The image demonstrates a cancer metastasis at 40x with beta1 staining (in red) in both blood vessels and tumour cells. Blue is Hoechst for nuclei. |
