Additional notes

Mice were perfused via heart with 10 ml PBS followed by 10 ml 4%PFA in PBS. Organs were left over night in 4% PFA/PBS at 4 C. Organs were washed in PBS and then transfered into a 30% succrose solution for cryoprotection and left over night at 4 C. Sections wer 10 um, dried and fixed in ice cold acetone for 10 min. Blocking was 30 min with 10 % normal goat serum.
I have tried antibody dilutions of 1:50 to 1:400 and found 1:50 to be working best.

Notes

We have used the antibody for immunostaining of frozen sections of mouse tumors and wounds.The best results was obtained with a 1:1000 dilution in TBS-BSA.Secondary ab: rabbit anti-rat 1:600(E468, DAKO), followed by HRP-streptavidin, 1:500(DAKO) and Nova red. It worked very well.
We also stained tissue prefixed with Stefani's fixative with good results.

Dilution notes

Primary ab: 1:1000 dilution in TBS-BSA.
Secondary ab: rabbit anti-rat 1:600(E468, DAKO)
Tert: ab: HRP-streptavidin, 1:500(DAKO)
