I stained some slides overnight. I seem to be getting a lot of background, I could increase my blocking step. I will try that and see if it cleans it up. I am staining human tissue.

Is there a paper that specifically uses this antibody in formalin fixed paraffin embedded tissue that I can reference.

only references we have available are those listed on the datasheet. Unfortunately, I am not able to access them to determine if there is any information regarding the use of this antibody in IHC-P. As I mentioned previously, if these modifications do not improve your results, I am happy to offer a replacement or credit.

We used Heat mediated retrieval at pH 9. The dilution of the antibody was 1/50, 1/100 and 1/200. Room temperature incubation for 30 minutes, with a polymer based detection. And then overnight at4C with the polymer based detection.

This is our routine IHC staining protocol for all clinical work. So this is where I started. There is staining but the pathologist feels that it is non specific. The data sheet lists Breast CA as a control, but what type of breast cancer. The pathologist also said that itshould stain basement membrane in normal epidermis....this is what we could not get good staining with.

Any help would be greatly appreciated.

Based on what you have already tested, I am not sure there are many more steps to be optimized. One consideration would be to extend the primary antibody incubation to overnight at 4oC. Longer incubation with less antibody tends to result in a cleaner, more specific signal. I am also happy to send out a new vial of antibody or issue a credit if you would prefer.

I cannot get this antibody to stain. I am using formalin fixed paraffin embedded tissue. The samples we are staining are skin, and then a TMA block with over thirty different types of tissue in it. I have tried ph6 citrate retrieval, ph9 Tris/EDTA retrieval and protienase K digestion. Any ideas? Thank you.

I am sorry that you have been experiencing difficulties with this antibody in IHC of paraffin embedded tissue. Heat mediated antigen retrieval using the Tris/EDTA pH 9.0 buffer is the recommended method.
