The Human Interferon gamma ELISA is to be used for the in-vitro quantitative determination of interferon gamma (Interferon gamma) in human serum, plasma, buffered solutions or cell culture medium. The assay will recognize both natural and recombinant human Interferon gamma.
Principal:
A monoclonal antibody specific for Interferon gamma has been coated onto the wells of the microtiter strips provided. During the first incubation, Interferon gamma present in the sample or standard and a monoclonal anti-Interferon gamma antibody conjugated to biotin are simultaneaously incubated. Following incubation unbound biotinylated anti-Interferon gamma is removed during a wash step. Streptavidin-HRP is added and binds to the biotinylated anti-Interferon gamma. After incubation and a wash step a substrate solution reactive with HRP is added to the wells. A coloured product is formed in proportion to the amount of Interferon gamma present in the sample. The reaction is terminated by addition of acid and absorbance is measured at 450 nm.

Mammalian Interferon gamma is mainly produced by T lymphocytes and NK cells. It is a pleiotropic cytokine involved in the regulation of nearly all phases of immune and inflammatory responses,including the activation, growth and differentiation of T cell, B cells, macrophages, NK cells and other cell types such as endothelial cells and fibroblasts. It has weak antiviral and antiproliferative activity, and poteniates the antiviral and anti tumor effects of IFN alpha / beta (type I interferon). It is upregulated by IL2, FGF basic, EGF and downregulated by vitamin D3 or DMN. Labile at pH 2.
