Mouse, Rat, Human Predicted to work with:
Synthetic peptide corresponding to Human JNK3 aa 300-400 conjugated to Keyhole Limpet Haemocyanin (KLH). (Peptide available as ab101295, ab101295)
This antibody gave a positive signal in U87mg whole cell lysate as well as the following tissue lysates: Human brain; Human spinal cord; Mouse hippocampus; Mouse cerebellum; Rat hippocampus; Rat cerebellum.
Storage instructionsShipped at 4°C. Store at +4°C short term (1-2 weeks). Upon delivery aliquot. Store at -20°C or -80°C. Avoid freeze / thaw cycle.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use a concentration of 1 µg/ml. Detects a band of approximately 48 kDa (predicted molecular weight: 52 kDa).
FunctionResponds to activation by environmental stress and pro-inflammatory cytokines by phosphorylating a number of transcription factors, primarily components of AP-1 such as c-Jun and ATF2 and thus regulates AP-1 transcriptional activity. Required for stress-induced neuronal apoptosis and the pathogenesis of glutamate excitotoxicity.
Tissue specificitySpecific to a subset of neurons in the nervous system. Present in the hippocampus and areas, cerebellum, striatum, brain stem, and weakly in the spinal cord. Very weak expression in testis and kidney.
Involvement in diseaseDefects in MAPK10 are a cause of epileptic encephalopathy Lennox-Gastaut type (EELG) [MIM:606369]. Epileptic encephalopathies of the Lennox-Gastaut group are childhood epileptic disorders characterized by severe psychomotor delay and seizures. Note=A chromosomal aberration involving MAPK10 has been found in a single patient. Translocation t(Y;4)(q11.2;q21) which causes MAPK10 truncation.
Sequence similaritiesBelongs to the protein kinase superfamily. CMGC Ser/Thr protein kinase family. MAP kinase subfamily. Contains 1 protein kinase domain.
DomainThe TXY motif contains the threonine and tyrosine residues whose phosphorylation activates the MAP kinases.
Post-translational modificationsDually phosphorylated on Thr-221 and Tyr-223, which activates the enzyme. Weakly autophosphorylated on threonine and tyrosine residues in vitro.

Predicted band size : 52 kDa Observed band size : 48 kDa (why is the actual band size different from the predicted?) Additional bands at : 34 kDa,52 kDa. We are unsure as to the identity of these extra bands. The predicted molecular weight of MAPK10 is 52 kDa (SwissProt), however we expect to observe a banding pattern at 48 kDa. Abcam welcomes customer feedback and would appreciate any comments regarding this product and the data presented above.
