    Recombinant fusion protein (N-terminal), corresponding to amino acids 103 - 584 of mouse Kaiso.

  • Positive control
    • 3T3/A31 cell lysate. Add 2.5µl of 2-mercaptoethanol/100µl of lysate and boil for 5 minutes to reduce the preparation. Load 20µg of reduced lysate per lane for minigels.
  • General notesFor maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

  • Application notesGSA: Use at an assay dependent dilution. Supershifts a specific methyl-CpG complex.
    ICC: Use at a concentration of 1 µg/ml for positive nuclear staining in NIH3T3 cells fixed with 100% methanol.
    IP: Use at an assay dependent dilution.
    WB: Use at a concentration of 0.5 - 2 µg/ml on RIPA lysates from 3T3/A31 cells (ECL). Detects a band of approximately 97 kDa (predicted molecular weight: 75 kDa).

    Not tested in other applications.
    Optimal dilutions/concentrations should be determined by the end user.
  • Target

    • FunctionTranscriptional regulator with bimodal DNA-binding specificity. Binds to methylated CpG dinucleotides in the consensus sequence 5'-CGCG-3' and also binds to the non-methylated consensus sequence 5'-CTGCNA-3'. Recruits the N-CoR repressor complex to promote histone deacetylation and the formation of repressive chromatin structures in target gene promoters. May contribute to the repression of target genes of the Wnt signaling pathway. May also activate transcription of a subset of target genes by the recruitment of CTNND2.
