Antibody purification kits

 Purification of antibodies from unwanted buffer constituents (e.g., BSA)

Commercially available antibodies often contain substances (e.g., BSA, glycine, tris, azide) that may interfere with applications such as antibody conjugation. Purify up to 500 µg of antibody easily using our kits that have been developed for antibody purification from 100 - 500 µL volumes:

These kits can also be used to purify antibodies from 100 - 500 µL of crude samples such as ascites fluid or serum. Antibodies purified using these kits are fully compatible with our conjugation kits, with the exception of our GOLD conjugation kits.

The method involves capture of the antibody on protein A, protein G or mouse specific resin and the removal of unwanted substances by a simple wash procedure using a standard microfuge. The purified product is then eluted and neutralized. This method cannot be used with samples containing relatively dilute antibody in large volumes e.g., tissue culture supernatant. The Mouse Antibody Purification Kit contains a proprietary affinity resin that has high affinity for mouse  IgG (independent of sub-type) and negligible binding to bovine IgG. The BSA removal kit is fully compatible with all antibody species and sub-types.


Antibody purification from tissue culture supernatant (TCS)

Our antibody TCS purification kits are designed to purify up to 5 mg of antibody from 10 - 50 mL of tissue culture supernatant:

The antibody is captured using the TCS resin and unwanted substances are removed by a simple wash procedure. The purified product is then eluted and neutralized.

The Antibody TCS purification kit ab109207 uses high affinity protein A agarose beads to purify IgG fractions from supernatants. The Mouse TCS purification kit ab128749 uses a proprietary affinity resin that selectively purifies mouse IgG from hybridoma supernatants without co-purifying bovine IgG.

Antibodies purified using these kits are fully compatible with our conjugation kits, with the exception of our GOLD conjugation kits.


Antibody purification from serum or ascites fluid

Our antibody serum purification kits are prepared by coupling highly purified protein A or G to agarose beads and can therefore be used to purify up to 20 mg of IgG from serum or ascites fluid:

The antibody is captured on the agarose beads and unwanted substances are removed by a simple wash procedure. The purified product is then eluted and neutralized. The volume of sample required will depend on the host species and is specified on the product datasheet.

Antibody concentration kit

Our antibody concentration kit enables quick and easy concentration of antibodies and proteins using a spin column:

In addition, if desired, the kit can also be used to reduce the concentration of low molecular weight buffer constituents often found in antibody preparations, such as azide, glycine or tris. Commercially available antibodies often contain substances (e.g. BSA, glycine, tris, azide) that interfere with your experiments.
 
Abcam's antibody purification kits quickly remove these contaminants. They can also be used to purify antibodies from crude samples such as ascites fluid or serum. The optimal volume of the sample to be purified is 100-500 µL and up to 500 µg of antibody can be purified in each run.

The method involves capture of the antibody on protein A, protein G or mouse specific resin and the removal of unwanted substances by a simple wash procedure, which is carried out in a standard microfuge. The purified product is then eluted and neutralized. This method cannot be used with samples containing relatively dilute antibody in large volumes (e.g. tissue culture supernatant).

Protein G resin has a high affinity for the Fc regions of IgG molecules from a variety of species (e.g. sheep, rat or goat). Protein A resin has a high affinity for the Fc regions of rabbit IgG molecules.

For purifying mouse antibodies from crude samples such as ascites the Mouse Antibody purification kit is recommended with a high affinity for mouse IgG molecules (independent of the sub-isotype) and negligible binding to Bovine IgG. The Mouse Antibody purification kit is based on a proprietary affinity resin and is not suitable for use with antibodies from other species.
