Description
Rabbit polyclonal to Kv1.6/RCK2 -
Host species
Rabbit -
Specificity
In human HEK293 cells ab21089 recognised a band of ~58 kDa, the predicted MW of Kv1.6/RCK2 (data not shown). This band was not blocked by the immunising peptide. Additional bands of 100 kDa, 25 kDa and 20 kDa were also recognised.

Target
-
Function
Mediates the voltage-dependent potassium ion permeability of excitable membranes. Assuming opened or closed conformations in response to the voltage difference across the membrane, the protein forms a potassium-selective channel through which potassium ions may pass in accordance with their electrochemical gradient. -
Sequence similarities
Belongs to the potassium channel family. A (Shaker) (TC 1.A.1.2) subfamily. Kv1.6/KCNA6 sub-subfamily. -
Domain
The N-terminus may be important in determining the rate of inactivation of the channel while the tail may play a role in modulation of channel activity and/or targeting of the channel to specific subcellular compartments.
The segment S4 is probably the voltage-sensor and is characterized by a series of positively charged amino acids at every third position. -

Predicted band size: 58 kDaab21089 recognises a band corresponding to the predicted MW of Kv1.6/RCK2 in mouse brain lysate (lane 1). Antibody binding to this band may be blocked using the immunising peptide, ab23431 (lane 2). The antibody also recognises additional proteins. These may be degradation/cleavage products of Kv1.6/RCK2, or they may represent non-specific binding by the antibody.

ICC/IF image of ab21089 stained human HEK (Human embryonic kidney) 293 cells. The cells were PFA fixed (10 min), permabilised in TBS-T (20 min) and incubated with the antibody (ab21089, 5µg/ml) for 1h at room temperature. 1%BSA / 10% normal goat serum / 0.3M glycine was used to quench autofluorescence and block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue).
