ICC/IF Use a concentration of 0.1 - 10 µg/ml. 0.1-1.0ug/mL (Perox), 1.0-10ug/mL (IF)
WB Use a concentration of 1 µg/ml. Predicted molecular weight: 71 kDa. 1µg/mL of ab99045 was sufficient for detection of Kv4.3 in 20µg of Rat brain lysate by colorimetric immunoblot analysis using goat anti-mouse IgG:HRP as the secondary antibody.
IP Use at an assay dependent concentration.
IHC-P Use a concentration of 0.1 - 1 µg/ml.
IHC-Fr Use a concentration of 0.1 - 1 µg/ml.
Flow Cyt Use 1µg for 106 cells. ab170190-Mouse monoclonal IgG1, is suitable for use as an isotype control with this antibody.

Target

  • FunctionPore-forming (alpha) subunit of voltage-gated rapidly inactivating A-type potassium channels. May contribute to I(To) current in heart and I(Sa) current in neurons. Channel properties are modulated by interactions with other alpha subunits and with regulatory subunits.
  • Tissue specificityHighly expressed in heart and brain, in particular in cortex, cerebellum, amygdala and caudate nucleus. Detected at lower levels in liver, skeletal muscle, kidney and pancreas. Isoform 1 predominates in most tissues. Isoform 1 and isoform 2 are detected at similar levels in brain, skeletal muscle and pancreas.
  • Sequence similaritiesBelongs to the potassium channel family. D (Shal) (TC 1.A.1.2) subfamily. Kv4.3/KCND3 sub-subfamily.

  • Overlay histogram showing SH-SY5Y cells stained with ab99045 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab99045, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in SH-SY5Y cells fixed with 80% methanol (5 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.
