L-FABP was isolated and purified from livers of Wistar strain rats as described in: Takashi, K. et al; Isolation and characterization of the three fractions (DE-I, DE-II and DE-III) of rat liver Z-protein and the complete primary structure of DE-II. Eur. J. Biochem. 1983; 136, 589-601. As I have limited access to journals, I hope the publication above will provide you with the information you are looking for. |

I'm trying to find some antibodies (monoclonal or polyclonal) which are specific enough to distinguish the various members of Fatty Acid Binding Proteins (FABP). I find some in your website, such as ab7847 and ab7366. However, I can't get enough information about their specificity. In my knowledge, there are at least eight members of FABP family in human, they are heart-FABP, liver-FABP,intestinal-FABP,adipocyte-FABP,psoriasis-FABP,gastrotropin, PMP2 and brain-FABP. In your data sheet, you descibed the antobodies as "no cross-reactivities with other FABP types have been detected". I want to know if that indicated that you have ckecked all the eight FABP members for the specificity. I'm looking forward to hearing from you. Thanks. |

Both of these antibodies to L-FABP (ab7847 and ab7366) have been tested for cross reactivity with H-FABP, I-FABP. L-FABP and I-FABP are the most similar FABP's of the eight members of the FABP family. So it is very unlikely that the antibodies cross react with other members of the FABP family. However, cross reactivity with other FABP proteins has not specifically been tested - I shall make this more clear on the datasheet for ab7847. |

ICC/IF image of ab7847 stained HepG2 cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab7847, 5µg/ml) overnight at +4ºC. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
All lanes : Anti-liver FABP antibody (ab7847) at 1/1000 dilution
Lane 1 : Whole tissue lysate prepared from pig liver
Lane 2 : Whole tissue lysate prepared from pig liver
Lysates/proteins at 10 µg per lane.
