Applications

Our Abpromise guarantee covers the use of ab57992 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 - 5 µg/ml.

This antibody has only been tested in WB against the recombinant fragment used as immunogen. We have no data on the detection of endogenous protein.

ICC/IF Use a concentration of 5 µg/ml.

  • Western blot against tagged recombinant protein immunogen using ab57992 LRG1 antibody at 1ug/ml. Predicted band size of immunogen is 60 kDa
  • ICC/IF image of ab57992 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab57992, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96879, DyLight® 488 goat anti-mouse IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Thank you for your reply.

I have had a look at all the anti-LRG1 antibodies we have to see if any of them might be able to pick up this difference in glycosylation.

The only one I would suggest might be worth trying is the ab63836. ...
