    Ligand-specific transporter trafficking between intracellular organelles (TGN) and the plasma membrane. Plays a role in autocrine regulation of cell growth mediated by growth regulators containing cell surface retention sequence binding (CRS). May act as a hyaluronan (HA) transporter, either mediating its uptake for catabolism within lymphatic endothelial cells themselves, or its transport into the lumen of afferent lymphatic vessels for subsequent re-uptake and degradation in lymph nodes.

  • ab33682 staining LYVE1 in mouse uterus tissue sections by Immunohistochemistry (IHC-P - Formalin/PFA-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 3% serum for 30 minutes at 20°C; antigen retrieval was by heat mediation in citrate buffer, pH 6.0. Samples were incubated with primary antibody (1/200 in PBS) for 12 hours at 20°C. A HRP-conjugated goat anti-rabbit IgG polyclonal (1/200) was used as the secondary antibody.

  • ab33682 staining LYVE1 in pig knee synovium tissue sections by Immunohistochemistry (IHC-Fr - frozen sections). Tissue was fixed with acetone and blocked with 1% BSA for 30 minutes at 22°C. Samples were incubated with the primary antibody (1/100) for 2 hours at 22°C. An Alexa Fluor® 488-conjugated goat anti-rabbit IgG monoclonal (1/200) was used as the secondary antibody. Propidium iodide was used to stain nuclei (543/633nm).

    Predicted band size : 35 kDa
    Additional bands at : 35 kDa (possible mature (processed) protein),42 kDa,80 kDa. We are unsure as to the identity of these extra bands.

    Exposure time : 20 minutes

    The LYVE-1 protein has a predicted molecular weight of 35 kDa. The first 23 amino acids of the LYVE-1 sequence act as a signal sequence, and when cleaved the protein has an expected molecular weight of 32 kDa. We believe that the band observed at 35 kDa represents the 32 kDa product.

  • ICC/IF image of ab33682 stained JEG3 cells. The cells were 4% formaldehyde fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody ab33682 at 5µg/ml overnight at +4°C. The secondary antibody (green) was DyLight® 488 goat anti- rabbit (ab96899) IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
