The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use a concentration of 5 µg/ml.
1/50 - 1/100.
1/100 - 1/500. Detects a band of approximately 59 kDa (predicted molecular weight: 59 kDa).
FunctionMay play a role in subcellular trafficking between the endoplasmic reticulum and Golgi apparatus through interactions with the Rho-type GTPases. Binding to the NS3 protein of dengue virus type 2 appears to subvert this activity into the alteration of the intracellular membrane structure associated with flaviviral replication.
Tissue specificityUbiquitously expressed in all tissues examined with high levels in the testis.
Sequence similaritiesBelongs to the protein kinase superfamily. Ser/Thr protein kinase family. Contains 1 protein kinase domain.
DomainThe protein kinase domain is predicted to be catalytically inactive.
Cellular localizationCytoplasm > cell cortex. Endomembrane system. Cell projection > lamellipodium. Colocalizes with activated RAC3 to endomembranes and at the cell periphery in lamellipodia.
Immunocytochemistry/ Immunofluorescence - Anti-MADM antibody - Aminoterminal end (ab37619)
ab37619 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody ab37619 at 5µg/ml overnight at +4°C. The secondary antibody (green) was DyLight® 488 goat anti- rabbit (ab96899) IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
