Application Abreviews Notes
  • Application notesICC/IF: Use at an assay dependent dilution (PMID 18694567).
WB: Use at a concentration of 2 µg/ml. Detects a band of approximately 49 kDa (predicted molecular weight: 49 kDa).

Not yet tested in other applications.

  • Target

    • FunctionPlays a central role during spermatogenesis by repressing transposable elements and prevent their mobilization, which is essential for the germline integrity. Acts via the piRNA metabolic process, which mediates the repression of transposable elements during meiosis by forming complexes composed of piRNAs and Piwi proteins and govern the methylation and subsequent repression of transposons. Its association with piP-bodies suggests a participation in the secondary piRNAs metabolic process. Required for localization of germ-cell factors to the meiotic nuage.
    • Tissue specificityTestis-specific. Expressed in various cancer cell lines, probably due to demethylation of its promoter.
    • Sequence similaritiesBelongs to the maelstrom family.
Contains 1 HMG box DNA-binding domain.
    • Cellular localizationCytoplasm. Nucleus. Component of the meiotic nuage, also named P granule, a germ-cell-specific organelle required to repress transposon during meiosis. Specifically localizes to piP-bodies, a subset of the nuage which contains secondary piRNAs.

    Anti-Mael antibody images

    • IHC image of Mael staining in Rat Normal Liver formalin fixed paraffin embedded tissue section, performed on a Leica Bond™ system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab28661, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

IRDye 680 Conjugated Goat Anti-Rabbit IgG (H+L) at 1/15000 dilution

      Performed under reducing conditions.

Predicted band size : 49 kDa
Observed band size : 49 kDa
Additional bands at : 25 kDa,37 kDa. We are unsure as to the identity of these extra bands.
