SpecificityAb33580 is highly specific to MAP 2a, b and c expressed in dendrites and cell bodies of neurons. It recognizes MAP2a and MAP2b (~280kD doublet) and MAP2c (70kD). It shows no crossreactivity with other MAPs, tau and tubulin.
The clone number has been updated from (0.N.395) to (AP18) both clone numbers name the same antibody clone.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use a concentration of 1 - 2 µg/ml. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.
Use a concentration of 1 - 2 µg/ml. Perform heat mediated antigen retrieval before commencing the IHC protocol.
Use a concentration of 1 - 2 µg/ml. Predicted molecular weight: 199 kDa.
Use at an assay dependent concentration.
FunctionThe exact function of MAP2 is unknown but MAPs may stabilize the microtubules against depolymerization. They also seem to have a stiffening effect on microtubules.
Sequence similaritiesContains 3 Tau/MAP repeats.
Post-translational modificationsPhosphorylated at serine residues in K-X-G-S motifs by MAP/microtubule affinity-regulating kinase (MARK1 or MARK2), causing detachment from microtubules, and their disassembly (By similarity). MAP2A/c is phosphorylated. Phosphorylated upon DNA damage, probably by ATM or ATR. Isoform MAP2c is phosphorylated by FYN at Tyr-67.
