Ab33580 is highly specific to MAP 2a, b and c expressed in dendrites and cell bodies of neurons. It recognizes MAP2a and MAP2b (~280kD doublet) and MAP2c (70kD). It shows no crossreactivity with other MAPs, tau and tubulin.

|
Recommended dilutions ICC/IF: Use at an assay dependent dilution. IHC-F: Use at a concentration of 1 - 2 µg/ml. Staining of formalin fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6 for 10-20 min., followed by cooling at room temperature for 20 min. WB: Use at a concentration of 1 - 2 µg/ml. Predicted molecular weight: 199 kDa.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
The expression of MAP2 is developmentally regulated and its multiple forms arise by alternative splicing of a single gene. MAP2 belongs to the microtubule associated protein family. The proteins of this family are thought to be involved in microtubule assembly, which is an essential step in neurogenesis. The products of similar genes in rat and mouse are neuron specific cytoskeletal proteins that are enriched in dentrites, implicating a role in determining and stabilizing dentritic shape during neuron development. A number of alternatively spliced variants encoding distinct isforms have been described.
