Our Abpromise guarantee covers the use of ab88482 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 - 5 µg/ml. Predicted molecular weight: 108 kDa.
ELISA Use at an assay dependent concentration.
ICC/IF Use a concentration of 10 µg/ml.

  • FunctionActivates the JUN N-terminal pathway through activation of the MAP kinase kinase MAP2K7. Acts synergistically with PRDX3 to regulate the activation of NF-kappa-B in the cytosol. This activation is kinase-dependent and involves activating the IKK complex, the IKBKB-containing complex that phosphorylates inhibitors of NF-kappa-B.
  • Tissue specificityExpressed in the adult brain, liver, placenta and pancreas, with expression strongest in the pancreas.
  • Sequence similaritiesBelongs to the protein kinase superfamily. STE Ser/Thr protein kinase family. MAP kinase kinase kinase subfamily.
    Contains 1 protein kinase domain.
  • Post-translational
Autophosphorylated on serine and threonine residues.
  • Cellular localizationCytoplasm. Membrane.

Anti-MAP3K13 antibody images

  • ICC/IF image of ab88482 stained Panc-1 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody ab88482 at 10µg/ml overnight at +4°C. The secondary antibody (pseudo-colored green) was Alexa Fluor® 488 goat anti- mouse (ab150117) IgG (H+L) preadsorbed, used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (pseudo-colored red) at a 1/200 dilution for 1h at room temperature. DAPI was used to stain the cell nuclei (pseudo-colored blue) at a concentration of 1.43µM for 1hour at room temperature.

  • Anti-MAP3K13 antibody (ab88482) at 5 µg/ml + Partial tagged recombinant MAP3K13 protein (immmunogen) at 0.2 µg

Predicted band size : 108 kDa
Observed band size : 37 kDa (why is the actual band size different from the predicted?)
Western blot against tagged recombinant protein immunogen. Predicted band size of immunogen is 37 kDa.
  • Anti-MAP3K13 antibody (ab88482) at 5 µg/ml + K-562 cell lysate at 50 µg

Predicted band size : 108 kDa
Observed band size : 120 kDa (why is the actual band size different from the predicted?)
