Applications

Our Abpromise guarantee covers the use of ab85725 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB 1/2000 - 1/10000. Predicted molecular weight: 49 kDa.
IP Use at 2-5 µg/mg of lysate.
IHC-P 1/500 - 1/2000. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

Target

  • FunctionMay function as a transcription factor with dual roles in transcription initiation and termination. Binds to two sites, ME1a1 and ME1a2, within the MYC promoter having greater affinity for the former. Also binds to multiple G/C-rich sites within the promoter of the Sp1 family of transcription factors.
  • Tissue specificityPresent in kidney, liver and brain. In the brain, highest levels are found in motor cortex and midfrontal cortex (at protein level).
  • Sequence similaritiesContains 6 C2H2-type zinc fingers.
  • Cellular localizationNucleus. In brains of Alzheimer disease patients, present in a plaque-like structures.

Anti-MAZ antibody images

  • Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human prostate carcinoma tissue labelling MAZ with ab85725 at 1/1000 (1µg/ml). Detection: DAB.
  • All lanes : Anti-MAZ antibody (ab85725) at 0.1 µg/ml

Lane 1 : HeLa cells lysate at 50 µg
Lane 2 : HeLa cells lysate at 15 µg
Lane 3 : HeLa cells lysate at 5 µg

    Developed using the ECL technique

Predicted band size : 49 kDa
Observed band size : 60 kDa (why is the actual band size different from the predicted?)
Additional bands at : 35 kDa,39 kDa. We are unsure as to the identity of these extra bands.

    Exposure time : 30 seconds
  • Detection of Human MAZ by Immunoprecipitation in HeLa cell lysate (1 mg for IP, 20% of IP loaded) using ab85725 at 3 ug/mg lysate from HeLa cells (lane 1). Lane 2 is Control IgG IP. Subsequent WB detection was performed using 1 ug/ml ab85725. Detection: Chemiluminescence with exposure times of 30 seconds.
