The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
1/500 - 1/2000. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
1/2000 - 1/10000. Detects a band of approximately 115 kDa (predicted molecular weight: 115 kDa).
Use at an assay dependent concentration. PubMed: 21980527
FunctionActs as component of the MCM2-7 complex (MCM complex) which is the putative replicative helicase essential for 'once per cell cycle' DNA replication initiation and elongation in eukaryotic cells. The active ATPase sites in the MCM2-7 ring are formed through the interaction surfaces of two neighboring subunits such that a critical structure of a conserved arginine finger motif is provided in trans relative to the ATP-binding site of the Walker A box of the adjacent subunit. The six ATPase active sites, however, are likely to contribute differentially to the complex helicase activity.
Sequence similaritiesBelongs to the MCM family. Contains 1 MCM domain.
Post-translational modificationsPhosphorylated upon DNA damage, probably by ATM or ATR.
Cellular localizationNucleus. Binds to chromatin during G1 and detach from it during S phase.
Immunohistochemistry (Frozen sections) - Anti-MCM6 antibody (ab3730)Image from Kim TM et al., PLoS One. 2011;6(9):e25679. Epub 2011 Sep 28. Fig 6.; doi:10.1371/journal.pone.0025679; September 28, 2011, PLoS ONE 6(9): e25679.
Immunofluorescence analysis of rat kidney tissue collected at 0 hours (shame) and 24 hours after ischemic injury, and embryonic rat kidney tissue from embryonic day 17 (E17d).
MCM6 (red) was stained using ab3730 at 1/1000 dilution. Nuclei were stained with DAPI (blue).
