| Application notes (see key) | Recommended dilutions WB: A recommended starting concentration for Western blots is 1:1000 when using colorimetric substrates such as BCIP/NBT, and 1:5000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for non-human samples. When used against the reduced protein, this antibody identifies bands at 58 Kd (the zymogen), but not the activated forms at 45 Kd, and a series of further cleaved active forms. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |

| Relevance | Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. However, the enzyme encoded by this gene is activated intracellularly by furin within the constitutive secretory pathway. Also in contrast to other MMP's, this enzyme cleaves alpha 1 proteinase inhibitor but weakly degrades structural proteins of the extracellular matrix.
MMP11, also known as stromelysin 3, is overexpressed in the stromal cells of invasive breast carcinomas but not in the stromal cells surrounding benign breast fibroadenomas. MMP11 is secreted in its proenzyme from and the zymogen has a mass of 64 kDa. The enzyme is rapidly activated to a 58 kDa form, a 45-47 kDa size, then to a 28 kDa final active form. MMP11 is not constitutively produced by most cell types. It has been found in lung fibroblasts and gingival fibroblasts when stimulated with EGF, as well as breast tumor and osteocarcinoma cell lines. |
