Application notes

WB: 1/1000 when using colorimetric substrates such as BCIP/NBT - 1/5000 chemiluminescent substrates. When used against the reduced protein, the antibody identifies bands at 72 kDa and 55 kDa as well as activation/breakdown product at 48 kDa. Often the MMP15 is fully activated, and the 48 kDa band is the dominant band seen. Predicted molecular weight: 76 kDa. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Endopeptidase that degrades various components of the extracellular matrix. May activate progelatinase A.

Tissue specificity

Appeared to be synthesized preferentially in liver, placenta, testis, colon and intestine. Substantial amounts are also detected in pancreas, kidney, lung, heart and skeletal muscle.

Sequence similarities

Belongs to the peptidase M10A family.
Contains 4 hemopexin-like domains.

Domain

The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme.

The precursor is cleaved by a furin endopeptidase.
