| Application notes (see key) | Recommended dilutions WB: Recommended starting dilution of 1/1000 when using colorimetric substrates such as BCIP/NBT and 1/5000 when using chemiluminescent substrates. Predicted molecular weight: 29 kDa. When used against the reduced protein, ab38997 identifies bands at about 28 kDa and 18 kDa (the pro form and active forms). Note: MMP7 does not appear to be produced by most normal quiescent cells, but treatment of many cell types with the phorbol ester TPA stimulates its production. Because of the low protein levels produced (pg/mL) concentration of cell culture media is often required to visualize the bands by Western Blotting. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |

| Relevance | Matrix Metalloproteinases (MMPs) belong to a family of proteases that are essential for the breakdown of extracellular matrix (ECM). They are thus important in apoptosis, tumor cell growth, invasion, and metastasis, as well as angiogenesis and wound healing. Most MMPs contain common domain structures that include a signal sequence, a propeptide, a catalytic domain, and a hemopexin like (Hpx) domain. MMP7, also known as Matrilysin, is responsible for hydrolyzing proteoglycans and ECM glycoproteins. It is expressed in normal cells, specifically glandular epithelial cells. MMP7, is thought to play an important role in the early steps of tumor progression. Expression of MMP2, MMP7 and MMP11 is greater in pancreatic carcinoma than in normal pancreas. |
