Specificity

Ab38990 recognises MMP8 in its carboxiterminal region. It does not recognize the smallest active form of MMP8, since the hemopexin domain is cleaved to produce this form. It also does not cross react with the other MMP family members.

Application notes

WB: Recommended starting dilution of 1/1000 when using colorimetric substrates such as BCIP/NBT and 1/5000 when using chemiluminescent substrates. Predicted molecular weight: 53 kDa. When used against the reduced protein, ab38990 identifies bands at 64 kDa, 58 kDa, and a series of further cleaved active forms. Note: MMP8 levels in quiescent cells and tissues are minimal and mitogen stimulation or protein concentration is often needed to visualize the bands. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Domain

The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme.

Cellular localization

Cytoplasmic granule. Secreted > extracellular space > extracellular matrix. Stored in intracellular granules.
