    Abcam’s MIP-1a Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of mouse MIP-1a in serum, plasma, and cell culture supernatants.

    This assay employs an antibody specific for mouse MIP-1a coated on a 96-well plate. Standards and samples are pipetted into the wells and MIP-1a present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse MIP-1a antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MIP-1a bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

I purchased the ELISA sandwich kit ab100726 for quantification of CCL3 in mice samples. The kit is optimized, according to the vendor, for detection of CCL3 in cell supernatant, serum or plasma.
When I tried to use the kit to measure CCL3 concentration in spinal cord homogenates, I found that serial dilutions of these homogenates do not return a proporcional ammount of CCL3, and therefore I assume that some secondary products are causing an interference with the kit detection ability.
