  • Overlay histogram showing subclass specificity of ab172333. HeLa cells were fixed with 80% methanol (-20ºC, 5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% serum / 0.3M glycine to block non-specific protein-protein interactions followed by the primary antibody (1μg/1x106) as indicated: ab16881 (Rat IgG1), ab90458 (Rat IgG2a), ab81461 (Rat IgG2b), ab50610 (Rat IgGc) for 30 min at 22ºC. The secondary antibody (ab172333 - Mouse monoclonal (2A 8F4) Anti-Rat IgG2a, heavy chain - Alexa Fluor® 647) was used at 1/3000 dilution for 30 min at 22ºC. Unlabelled (blue line) and secondary only (gold line) samples were also used as negative controls. Acquisition of >5,000 events were collected using a solid-state 25mW red diode laser (635nm) and 675/30 bandpass filter.
