The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use a concentration of 5 µg/ml.
Use a concentration of 1 - 3 µg/ml. Detects a band of approximately 75 kDa (predicted molecular weight: 77 kDa).
FunctionMay play a role in tight junctions biogenesis and in the establishment of cell polarity in epithelial cells. May modulate SC6A1/GAT1-mediated GABA uptake by stabilizing the transporter. Required for localization of EZR to the apical membrane of parietal cells and may play a role in the dynamic remodeling of the apical cytoskeleton.
Tissue specificityExpressed in the retina (at protein level).
Sequence similaritiesBelongs to the MAGUK family. Contains 1 guanylate kinase-like domain. Contains 2 L27 domains. Contains 1 PDZ (DHR) domain. Contains 1 SH3 domain.
DomainThe L27 domain 1 functions in targeting to the tight junctions by binding to INADL. The PDZ domain binds to the C-terminus of SC6A1.
Cellular localizationCell membrane. Endomembrane system. Cell junction > tight junction. Localized to the tight junctions of epithelial cells and a subset of intracellular vesicles. In the retina, detected at the outer limiting membrane (OLM), apical to the adherens junction (AJ), where it colocalizes with CRB1. Colocalizes with MPP1 in the retina at the outer limiting membrane (OLM). Localized to the Purkinje cell body and axon.
ab20511 at 5ug/ml staining MPP5 in human skeletal tissue section by Immunohistochemistry (Formalin/PFA fixed paraffin-embedded sections). Tissue underwent antigen retrieval in steam with citrate buffer (pH 6.0). The AP-staining procedure was used for detection.
