ab123455 mtTFA (TFAM) Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human mtTFA in cell and tissue lysates. The assay employs an antibody specific for the Human mtTFA coated onto well plate strips.
Standards and samples are pipetted into the wells and analyte present in the sample is bound to the wells by the immobilized antibody. The wells are washed and an anti- mtTFA primary detector antibody is added. After washing away unbound primary detector antibody, HRP-conjugated secondary detector antibody specific for the primary detector antibody is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of analyte bound. The developing blue color is measured at 600 nm.
Optionally the reaction can be stopped by adding hydrochloric acid which changes the color from blue to yellow and the intensity can be measured at 450 nm.
Notes
Store all components at 4°C. This kit is stable for at least 6 months from receipt. After reconstitution the standard should be stored at -80°C. Unused microplate strips should be returned to the pouch containing the desiccant and resealed.

Function
Binds to the mitochondrial light strand promoter and functions in mitochondrial transcription regulation. Required for accurate and efficient promoter recognition by the mitochondrial RNA polymerase. Promotes transcription initiation from the HSP1 and the light strand promoter by binding immediately upstream of transcriptional start sites. Is able to unwind and bend DNA. Required for maintenance of normal levels of mitochondrial DNA. May play a role in organizing and compacting mitochondrial DNA. target DNA. Interacts with TFB1M and TFB2M.
