  • Function
    RNA binding protein that regulates the expression of target mRNAs at the translation level. Regulates expression of the NOTCH1 antagonist NUMB. Binds RNA containing the sequence 5'-GUUAGUUAGUUAGUU-3' and other sequences containing the pattern 5'-[GA]U(1-3)AGU-3'. May play a role in the proliferation and maintenance of stem cells in the central nervous system.

Anti-Musashi 1 / Msi1 antibody [EP1302] images

  • Immunocytochemistry/Immunofluorescence analysis of Neuro-2a (mouse neuroblastoma) labelling Musashi 1 with purified ab52865 at 1/500. Cells were fixed with 4% Paraformaldehyde and permeabilised by 0.1% tritonX-100. An Alexa Fluor® 488-conjugated goat anti-rabbit IgG (1/1000) was used as the secondary antibody (Ab150077). Nuclei counterstained with DAPI (blue).

  • Immunohistochemistical detection (on formaldehyde/PFA-fixed paraffin-embedded sections) of Musashi 1 / Msi1 antibody [EP1302] (ab52865) on Quail Tissue sections (embryo d5/6 Brain stem T/S). Antigen retrieval step: Heat mediated. Blocking step: 1% BSA for 10 mins at RT. Primary Antibody ab52865 incubated at 1/300 for 2 hours at RT. Secondary Antibody: Biotin labelled goat anti rabbit IgG (1/300).
  • Flow Cyt image of Musashi1 (ab52865)using Accutase digested single cell suspension of hESC. the cells were fixed adn permeabilized. The cells were incubated with ab52865 (1/20 using Prem/wash solution) for 30 mins at 23°C.

  • Musashi 1 / Msi1 was immunoprecipitated using 0.5mg SHSY5Y whole cell extract, 10µg of Rabbit monoclonal to Musashi 1 / Msi1 and 50µl of protein G magnetic beads (+). No antibody was added to the control (-).
    The antibody was incubated under agitation with Protein G beads for 10min, SHSY5Y whole cell extract lysate diluted in RIPA buffer was added to each sample and incubated for a further 10min under agitation.
    Proteins were eluted by addition of 40µl SDS loading buffer and incubated for 10min at 70oC; 10µl of each sample was separated on a SDS PAGE gel, transferred to a nitrocellulose membrane, blocked with 5% BSA and probed with ab52865.
    Secondary: Mouse monoclonal [SB62a] Secondary Antibody to Rabbit IgG light chain (HRP) (ab99697).
    Band: 39kDa: Musashi 1 / Msi1; non specific - 29kDa: We are unsure as to the identity of this extra band.
