|IHC-P||Use a concentration of 2 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.|
|WB||Use a concentration of 1 µg/ml. Detects a band of approximately 35 kDa.Can be blocked with Human MyD88 peptide (ab7882).
Use a modified RIPA lysis buffer: 150 mM sodium chloride, 50 mM Tris-HCl, pH 7.4, 1 mM ethylenediaminetetraacetic acid, 1 mM phenylmethylsulfonyl fluoride, 1% Triton X-100, 1% sodium deoxycholic acid, 0.1% sodium dodecylsulfate, 5 µg/ml of aprotinin, 5 µg/ml of leupeptin). Remove cell debris by centrifugation. Boil the lysate for 5 min in 1 x SDS sample buffer (50 mM Tris-HCl pH 6.8, 12.5% glycerol, 1% sodium dodecylsulfate, 0.01% bromophenol blue) containing 5% ß-mercaptoethanol). Block the membrane with 5% non-fat dry milk in TBS overnight at 4°C or 2 hours at room temperature.

ab2068 staining MyD88 in Human sigma / diverticulitis tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections).
