    NanO2 probe (ab172912) is a cell-penetrating phosphorescence-based O2 sensing probes enable direct high-resolution 2D and 3D mapping and quantitation of (intra)cellular O2. NanO2 probe (ab172912) and MM2 probe (ab172913), offer a simple and convenient approach to the direct real-time live-cell imaging in-vitro, in-vivo, and for 3D cultures on standard fluorescent microscopes (wide-field, confocal or two-photon.

    Number of Assays: 10 - 100 assays
    Number of tests vary depending on platform used in the assay. The probe contains sufficient material for either 100 assays in a standard 96 well plate or 10 assays in an imaging mini dish 35mm.

    Probe Preparation and Cell Loading
    Probe is supplied as 100 µg aliquots in plastic vials.

    1. Add 100 µL of water or medium to the vial and vortex for 30 seconds to produce 1 mg/mL stock solution of the probe.

    2. For use with cultured cells, spheroids or tissue slices dilute probe stock 1:100 with medium (10 µg/mL working concentration) and add this solution to the cells.

    Typical volumes:

    • 100 µL per well for standard 96-well plates
    • 1 mL for imaging mini-dishes or 200 µL for mini-dishes with 4-well inserts.

    3. Incubate at 37°C in a CO2 incubator for 3-24h, then wash the cells 1-2 times with fresh medium and conduct optical measurements.
    Depending on the cell type and application, optimal probe concentration (typical range 2- 20 µg/mL) and loading time may vary.

    Measurement Conditions

    • Excitation wavelenghts: 390-405 nm (optimal), or 505, 540 nm (less efficient)
    • Emission wavelengths: 640-670 nm (maximum 650 nm)
    • Lifetime range (37°C): 23 µsecond at 21% O2 (209 µM dissolved O2), 65 µsecond at 0% O2

    Temperature and gas equilibration of samples during the measurement is important since the probe is sensitive to both O2 and T.

    Handling & Storage

    • Undiluted probe should be stored in the dark at +4°C.
    • Diluted probe should be used on the same day. Stock can be stored for 1-2 weeks at +4°C.
    • Protect the probe and loaded cells from light. Stable under ambient light, however exposure to high-energy excitation light should be minimised.
