    Unsuitable for: IHC
  • Species reactivity
    Reacts with: Mouse, Human
    Predicted to work with: Rat
  • Immunogen

Synthetic peptide corresponding to Human NANOS2 (C terminal). 13 amino acid peptide near the carboxy terminus of the human NANOS2.

  • Positive control
    • SK-N-SH cell lysate, mouse brain, mouse spleen, A20, 3T3(NIH)
  • General notes

Previous batches of ab70000 have been successfully used in IHC-P on testis tissue sections (please see Kusz KM et al., 2009). However, when tested on human brain tissue sections, ab70000 gave negative results in IHC-P (data not shown). We do not guarantee the current batch of ab70000 to work in IHC.

Our Abpromise guarantee covers the use of ab70000 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 - 2 µg/ml. Detects a band of approximately 15 kDa (predicted molecular weight: 15 kDa).
  • Application notesIs unsuitable for IHC.
  • Target

    • FunctionPlays a key role in the sexual differentiation of germ cells by promoting the male fate but suppressing the female fate. Represses the female fate pathways by suppressing meiosis, which in turn results in the promotion of the male fate. Maintains the suppression of meiosis by preventing STRA8 expression, which is required for premeiotic DNA replication, after CYP26B1 is decreased. Regulates the localization of the CCR4-NOT deadenylation complex to P-bodies and plays a role in recruiting the complex to trigger the degradation of mRNAs involved in meiosis. Required for the maintenance of the spermatogonial stem cell population. Not essential for the assembly of P-bodies but is required for the maintenance of their normal state.
    • Tissue specificityTestis and ovary. Expression found in several spermatogenic stages: in cells on the periphery of the tubules which could correspond to spermatogonia, in spermatocytes and in round spermatids (at protein level).
    • Sequence similaritiesBelongs to the nanos family.
      Contains 1 nanos-type zinc finger.
    • Developmental stageFetal ovary and fetal testis. Present in all germ cells of seminiferous tubules of the 24-week fetus (at protein level).
    • DomainThe Nanos-type zinc finger is composed of two C2HC motifs, each motif binding one molecule of zinc. It is essential for the translation repression activity of the protein.
    • Cellular localizationCytoplasm. Cytoplasm > P-body. Cytoplasm > perinuclear region. Localizes at P-bodies during gonocyte development (By similarity). More abundant in perinuclear region of the cytoplasm of the germ cells of the adult testis.
