Anti-Nebulin antibody [NB2] (ab11099)

Overview

  • Product nameAnti-Nebulin antibody [NB2]
    See all Nebulin primary antibodies
  • Description
    Mouse monoclonal [NB2] to Nebulin
  • SpecificityDoes not cross react with titin. Monoclonal Anti-Nebulin reacts only with skeletal muscle, not with smooth muscle or different non-muscle tissues and cultured cells. The following is a summary of the antibody reactivity on various muscles in immunofluorescence using frozen section (a), isolated myofibrils (b), or immunoblotting (c): Fish skeletal muscle (a) (Torpedo spec.) + Toad skeletal muscle (a) (Bufo marinus) + Salamander skeletal muscle (c) (Ambystoma tigrina) + Salamander heart muscle (a) - Alligator skeletal muscle (a)(Alligator mississippiensis) + Pigeon skeletal muscle (b) (c) + Chicken skeletal muscle (b) (c) + Chicken heart muscle (c) - Chicken gizzard smooth muscle (c) - Mouse skeletal muscle (a) + Rat skeletal muscle (b) (c) + Rat heart muscle (a) - Rat rhabdomyosarcoma (a) + Rabbit skeletal muscle (b) + Bovine heart muscle (a) - Pig skeletal muscle (c) + Human skeletal muscle (a) +

  • Application notesIF: 1/400.
WB: Use at an assay dependent dilution. Detects a band of approximately 500 kDa.

By indirect immunofluorescence, the antibody shows a typical striated staining pattern of frozen sections of chicken skeletal muscle. It stains within the I-band and shows a decoration line at a distance of 0.22 µm from the z-line in electron micrographs.

    Not tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
  • Target

    • FunctionThis giant muscle protein may be involved in maintaining the structural integrity of sarcomeres and the membrane system associated with the myofibrils. Bind and stabilize F-actin.
    • Tissue specificityMuscle specific. Located in the thin filament of striated muscle.
    • Involvement in diseaseDefects in NEB are the cause of nemaline myopathy type 2 (NEM2) [MIM:256030]. A form of nemaline myopathy. Nemaline myopathies are muscular disorders characterized by muscle weakness of varying severity and onset, and abnormal thread-or rod-like structures in muscle fibers on histologic examination.
    • Sequence similaritiesContains 178 nebulin repeats.
      Contains 1 SH3 domain.
    • Cellular localizationCytoplasm > myofibril > sarcomere. Cytoplasm > cytoskeleton.
