The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Application notesTested in peptide ELISA: antibody detection limit dilution 1:16,000.
IHC-P: Use at a concentration of 5 µg/ml.
Western Blot: preliminary experiments give ~60kDa band in human liver and brain lysates at 1ug/ml. However predicted band size = 44kDa (NP_078884)
Not tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
FunctionInvolved in base excision repair of DNA damaged by oxidation or by mutagenic agents. Acts as DNA glycosylase that recognizes and removes damaged bases. Has a preference for oxidized pyrimidines, such as thymine glycol, formamidopyrimidine (Fapy) and 5-hydroxyuracil. Has marginal activity towards 8-oxoguanine. Has AP (apurinic/apyrimidinic) lyase activity and introduces nicks in the DNA strand. Cleaves the DNA backbone by beta-delta elimination to generate a single-strand break at the site of the removed base with both 3'- and 5'-phosphates. Has DNA glycosylase/lyase activity towards mismatched uracil and thymine, in particular in U:C and T:C mismatches.
ab14753 at 5ug/ml staining NEIL1 in human placental tissue section by Immunohistochemistry (Formalin/PFA fixed paraffin-embedded sections). Tissue underwent antigen retrieval in steam with citrate buffer (pH 6.0). A AP-staining procedure was used for detection.
