Applications

Our Abpromise guarantee covers the use of ab72908 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
ICC/IF Use a concentration of 5 µg/ml.
WB Use a concentration of 1 µg/ml. Detects a band of approximately 140 kDa (predicted molecular weight: 135 kDa).

Target

  • FunctionSeems to play a role in the development or function of the kidney glomerular filtration barrier. Regulates glomerular vascular permeability. May anchor the podocyte slit diaphragm to the actin cytoskeleton. Plays a role in skeletal muscle formation through regulation of myoblast fusion.
  • Tissue specificitySpecifically expressed in podocytes of kidney glomeruli.
  • Involvement in diseaseDefects in NPHS1 are the cause of nephrotic syndrome type 1 (NPHS1) [MIM:256300]; also known as Finnish congenital nephrosis (CNF). A renal disease characterized clinically by proteinuria, hypoalbuminemia, hyperlipidemia, and edema. Kidney biopsies show non-specific histologic changes such as focal segmental glomerulosclerosis and diffuse mesangial proliferation. Some affected individuals have an inherited steroid-resistant form and progress to end-stage renal failure.
  • Sequence similaritiesBelongs to the immunoglobulin superfamily.
    Contains 1 fibronectin type-III domain.
    Contains 8 Ig-like C2-type (immunoglobulin-like) domains.
  • Developmental stageIn 23-week-old embryo found in epithelial podocytes of the periphery of mature and developing glomeruli.
  • Post-translational
    modifications
    Phosphorylated on tyrosine residues.
  • Cellular localizationCell membrane. Predominantly located at podocyte slit diaphragm between podocyte foot processes. Also associated with podocyte apical plasma membrane.

  • Anti-Nephrin antibody (ab72908) at 1 µg/ml + Human kidney tissue lysate - total protein (ab30203) at 10 µg

    Secondary
Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (HRP), pre-adsorbed at 1/5000 dilution
    Developed using the ECL technique

    Performed under reducing conditions.

Predicted band size : 135 kDa
Observed band size : 140 kDa (why is the actual band size different from the predicted?)
Additional bands at : 44 kDa,47 kDa. We are unsure as to the identity of these extra bands.

    Exposure time : 2 minutes
  • ICC/IF image of ab72908 stained Hek293 cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab72908, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96879, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM. This antibody also gave a positive result in 4% PFA fixed (10 min) HeLa cells at 5µg/ml, and in 100% methanol fixed (5 min) HeLa and Hek293 cells at 5µg/ml.
