The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use a concentration of 4 - 6 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
Use a concentration of 0.1 - 0.3 µg/ml. Detects a band of approximately 15 kDa (predicted molecular weight: 8 kDa). An approximately 15 kDa band is observed in Human Brain (Cerebral Cortex and Hippocampus), and in Mouse Brain and Rat Brain lysates. There is also an additional 16kDa band. The observed molecular weights correspond to earlier findings in the literature with different antibodies.
FunctionActs as a "third messenger" substrate of protein kinase C-mediated molecular cascades during synaptic development and remodeling. Binds to calmodulin in the absence of calcium.
Tissue specificityIn the cerebral cortex, found in the cell bodies of neurons in layers II-VI, and in apical and basal dendrites of pyramidal neurons. Is not found in the dendrites in patients with Alzheimer disease.
Sequence similaritiesBelongs to the neurogranin family. Contains 1 collagen-like domain. Contains 1 IQ domain.
