Recommended dilutions
ICC/IF: 1/200. Fix with paraformaldehyde (see Abreview).
IHC-P: Use at an assay dependent concentration.
IHC-Fr: 1/500.
IHC-FoFr: 1/200 (PMID 19200230).
WB: Use at a concentration of 1 µg/ml. Detects a band of approximately 35 kDa (predicted molecular weight: 32 kDa).
Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Cellular localization
|
Nuclear and cytoplasmic. The NLS contained in the bHLH domain could be masked in the native form and translocation to the nucleus could be mediated by interaction either with class E bHLH partner protein or with NKX2-2.

Relevance
|
Olig2 (oligodendrocyte transcription factor 2) is required for oligodendrocyte and motor neuron specification in the spinal cord, as well as for the development of somatic motor neurons in the hindbrain. It cooperates with OLIG1 to establish the pMN domain of the embryonic neural tube. It is an antagonist of V2 interneuron and of NKX2-2-induced V3 interneuron development.

 Immunohistochemistry (Frozen sections) - Olig2 antibody (ab33427)
ab33427 staining E14.5 mouse brain tissue sections by IHC-Fr. Sections were PFA fixed and permeabilized in 0.1% Triton X-100 prior to blocking in 10% serum for 30 minutes at 20°C. The primary antibody was diluted 1/500 and incubated with the sample for 16 hours at 4°C. A Cy3® conjugated donkey anti-rabbit IgG antibody, diluted 1/200, was used as the secondary.
