    Abcam’s Oxytocin in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of Oxytocin in Tissue Culture media, Human Serum, Saliva, Cerebrospinal Fluid and Urine.

    A goat anti-rabbit IgG antibody has been precoated onto 96-well plates. Standards or test samples are added to the wells, along with an alkaline phosphatase (AP) conjugated-Oxytocin antigen and a polyclonal rabbit antibody specific to Oxytocin. After incubation the excess reagents are washed away. pNpp substrate is added and after a short incubation the enzyme reaction is stopped and the yellow color generated is read at 405 nm. The intensity of the yellow coloration is inversely proportional to the amount of Oxytocin captured in the plate.

    Oxytocin is a neurohypophysial peptide which is produced in the paraventricular nuclei of the hypothalamus and stored in the posterior pituitary. The molecule consists of nine amino acids linked with a [1-6] disulfide bond and a semi-flexible carboxyamidated tail. A hormone once thought to be limited to female smooth muscle reproductive physiology, more current findings have determined that oxytocin also functions as a neurotransmitter, may be involved in neuropsychiatric disorders, social/sexual behavior and is important in male reproductive physiology. Oxytocin and the related neurohypophysial peptide, Arg-Vasopressin, maintain renal water and sodium balance.  

    Highly conserved across species boundaries, oxytocin-like neurohypophysial peptides are substituted primarily at residues 4 and/or 8. In the Oxytocin-like peptide, mesotocin, a common peptide found in some fishes, reptiles, amphibians, marsupials and nonmammalian tetrapods, the leucine at residue 8 is substituted for isoleucine. Acting in classical endocrine fashion, Oxytocin elicits regulatory effects by binding specific cell surface receptors which in turn initiate a secondary intracellular response cascade via a phosphoinositide signaling pathway.

We used ab133050 in order to quantify the amount of oxytocin in frozen rat brain tissue samples approx. 1mm2.
Immediately before running the assay we homogenized the samples in the ice cold complete lysis buffer recommended by abcam. The samples were then centrifuged once and loaded directly to the assay plate.
We were pleased to find that oxytocin was detected within the range of the standard curve.
Had a minor issue with the washes, the amount of washing buffer recommended per well was excessive and we reduced it from 400 to 350 microliter. Otherwise the instructions are clear and the kit is user-friendly.

Oxytocin elisa kit

The elisa kit is very friendly and works fine, but we had problems with the sample extraction procedure.
The extraction protocol was not clear for researchers that it’s the first time they met this procedure. For example you wrote that the volume of the washes with 0.1% TFA is between 10-25 ml (?!). When we evaporated the 3ml elution solution we found out that its take almost two days (?!).
We looked for "protein researchers" and they guided us how to do it properly.
Another comment when we washed the plate with 400µl as mention in the elisa procedure (13.8) the wash buffer spilled out of the walls, so we washed the wells with 380 µl.
Apart of that, the kit is very good. The standard curve we received was ok (attached curve fit that was calculated with 4 parameters logistic). Nonetheless, our samples were from rat serum and in many samples we received signals lower than the lowest standard (lower than 15.6 pg/ml). We understand that the oxytocin concentration in these samples is very low.
We tried to measure 3 samples as serum without the extraction procedure and we received at these samples signals lower than the lowest standard while when we measured them after extraction we received a detectable signal. Therefore, we understand that the extraction step is crucial for the detection.

Oxytocin Kit - ab133050

I conducted the analyzes according to the protocol suggested in the manual using the dilution of the sample (mouse serum) at 1: 5.
My only criticism is due to the complexity of the analysis of results (obtaining curve and calculations)
