Description

Mouse monoclonal [p150-1] to p150 CAF1

Specificity

This antibody is specific for p150, the subunit of CAF1 that directly contacts PCNA. Immunostaining with anti-CAF1 antibody localizes the protein to sites of DNA replication in the cell.

Target

Relevance

Chromatin assembly factor 1 (CAF1) is a member of the histone chaperone family. It is required for inheritance of epigenetically determined chromosomal states, and in vitro, it promotes the assembly of chromatin during DNA replication and DNA repair. CAF1 is a complex of three subunits; p150, p60, and p48. This complex is localized to the nucleus of human cells. Both the composition and the biochemical activity of CAF1 are conserved among human, yeast, and Drosophila.

ab24746 staining cultured human HeLa cells by ICC/IF.  Cells were PFA fixed and permeabilized in 0.5% Triton X100 prior to blocking in 5% BSA for 1 hour at 20°C.  The primary antibody was diluted 1/100 and incubated with the sample for 1 hour at 20°C.  A Cy3® conjugated donkey anti-mouse antibody diluted 1/400 was used as the secondary.
