Hi, I appreciate your prompt response. The samples are human cell extracts. I have not attempted other concentrations since the product sheet suggested the dilution of 1:20,000. Normally, I try to "optimize" the antibody concentration if there is no recommendations. Since this antibody had been tested and verified, I only tested the one concentration (1:10,000). I informed my supervisor of the situation and mentioned that Abcam is willing to send another lot. Please let me know if the situation is true. Thanks for all your help!

Hi, I appreciate your help and follow-up. I have attached the scanned film of the two western blots I performed. As I explained in our conversation, the p16 antibody from a different vendor gives a faint positive band, but the high background interferes with the signal. Therefore, I am interested in finding another antibody with better detection. The purchase order number is PO 51122A0093 and order number is 1009543. Have you tried increasing the antibody concentration? When we spoke you had mentioned 1:10,000 over night, have you tried 1:5,000, 1:1,000 or even 1:500? I have thought that it may be that theremay be a cross reaction between theantibody and your block.Have you tried to change your block to BSA or gelatin? I hope this information is helpful to you.
