Recommended dilutions ICC/IF: Use at a concentration of 5 µg/ml. WB: Use at a concentration of 0.2 (endogenous) - 1 (recombinant) µg/ml. Detects a band of approximately 35 kDa (predicted molecular weight: 28 kDa).
Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
RAB GTPases are involved in the docking of transport vesicles with their cognate target membranes. The association of RAB proteins with membranes is enabled by their doubly geranylgeranylated, hydrophobic C termini. RAB9 GTPase is required for the stimulation of mannose 6-phosphate receptor transport from endosomes to the trans-Golgi network. p40 is thought to be a RAB9 effector protein with kelch motifs.

Detection of recombinant human p40 using ab12109
Performed under reducing conditions.
Predicted band size : 28 kDa Additional bands at : 32 kDa (possible cross reactivity),47 kDa (possible cross reactivity),49 kDa (possible cross reactivity),60 kDa (possible cross reactivity).
ab12109 recognises p40 in Western Blots.
A: 293 HEK cells were transfected with negative control cDNA (pCDNA3 vector) or human p40 cDNA.
B: negative control cDNA (pCDNA3 vector) or human p40 cDNA were in vitro translated in rabbit reticulocyte lysate.
Blots were subjected to ab12109 diluted 1/2500. Exposure was for 1 minute.
A band corresponding to the expected MW of p40 was detected (arrow) in all lanes except negative control lanes. Exposure time: 1 min

Immunocytochemistry/ Immunofluorescence - p40 antibody (ab12109)
ICC/IF image of ab12109 stained human HeLa cells. The cells were methanol fixed (5 min) and incubated with the antibody (ab12109, 5µg/ml) for 1h at room temperature. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Image-iTTM FX Signal Enhancer was used as the primary blocking agent, 5% BSA (in TBS-T) was used for all other blocking steps. DAPI was used to stain the cell nuclei (blue). Alexa Fluor® 594 WGA was used to label plasma membranes (red).
