Our Abpromise guarantee covers the use of ab4060 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
|IHC-P||1/50 - 1/100. Perform heat mediated antigen retrieval before commencing with IHC staining protocol. 30mins at RT (ABC method). 10mM citrate buffer pH 6.0 is recommended for antigen retrieval step.|
|IHC-Fr||Use at an assay dependent concentration.|
ab4060 at 1/100 staining human breast tissue sections by IHC-P. The tissue was formaldehyde fixed and a heat mediated antigen retrieval step was performed. The tissue was blocked and then incubated with the antibody for 12 hours at 4°C. An HRP conjugated goat anti-rabbit antibody was used as the secondary.
ab4060 at a 1/100 dilution staining p53 in human skin tissue (Bowen's disease) by Immunohistochemistry (Formalin fixed, paraffin embedded) incubated for 20 hours at 4°C. Heat mediated antigen retrieval step performed using citrate buffer pH 6.0, microwave 10 minutes. Permeabilized using Tris-Buffered Saline Tween-20. Blocked using 2% BSA for 1 hour at 22°C. Secondary used at 1/200 dilution Goat polyclonal to anti-Rabbit FITC (IgG H&L)(green) (ab6717). The nuclei are counterstained with propidium iodide (red).
