ICC/IF image of ab3125 stained PC12 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab3125, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

ab3125 staining p75 NGF Receptor in Human cholinergic neurons by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded tissue sections). The sections were fixed in formalin and subjected to heat-mediated antigen retrieval in sodium citrate buffer prior to blocking with 2.5% serum for 1 hour 30 minutes at 21°C. The primary antibody was diluted 1/250 in normal horse serum blocking buffer and incubated with the sample for 65 hours at 4°C.

Samples were then permeabilized using 0.25% Triton X-100, blocked for 1 hour at room temperature and then incubated with ab3125 at a 1/100 dilution for 18 hours at 4°C. The secondary used was an Alexa-Fluor 594 (red) conjugated donkey anti-mouse polyclonal used at a 1/100 dilution. (Green) tumor cells, (blue) DAPI.
