Anti-p75 NGF Receptor antibody images

  • ICC/IF image of ab38335 stained rat PC12 cells. The cells were methanol fixed (5 min), permabilised in PBS-T (20 min) and incubated with the antibody (ab38335, 1µg/ml) for 1h at room temperature. 1%BSA / 10% normal goat serum / 0.3M glycine was used to block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue).

  • ab38335 (1/100) staining p75 NGF receptor in C57B/J mouse vagus nerve. Frozen cryosections were fixed with paraformaldehyde, permeabilised in 0.05% Triton X-100/PBS and blocked with 5%BSA/0.5% goat serum/PBST for 1 hour. for further experimental details please refer to Abreview.

    Exposure time : 2 minutes

    This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 2% Bovine Serum Albumin before being incubated with ab38335 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution ab133406

    Additional bands at : 50 kDa,69 kDa. We are unsure as to the identity of these extra bands.

    p75 NGF Receptor is known to run at 75 kDa due to potential glycosolation and phosphorylation sites. We believe that the bands observed at 70 kDa and 69 kDa represent p75 NGF Receptor. The 69 kDa band may correspond to the non-phosphorylated form of the protein.

  • ab38335 staining p75 in HeLa cells treated with Leptomycin B (ethanol solution) (ab120501), by ICC/IF. Increase in p75 expression in the nucleous and cytoplasm correlates with increased concentration of Leptomycin B (ethanol solution), as described in literature.
    The cells were incubated at 37°C for 2h in media containing different concentrations of ab120501 (Leptomycin B (ethanol solution)) in DMSO, fixed with 100% methanol for 5 minutes at -20°C and blocked with PBS containing 10% goat serum, 0.3 M glycine, 1% BSA and 0.1% tween for 2h at room temperature. Staining of the treated cells with ab38335 (1 µg/ml) was performed overnight at 4°C in PBS containing 1% BSA and 0.1% tween. A DyLight 488 goat anti-rabbit polyclonal antibody (ab96899) at 1/250 dilution was used as the secondary antibody. Nuclei were counterstained with DAPI and are shown in blue.
