I have a question about imunohistochemistry using these antibodies. Some of them (ab11427/ ab3267/ ab7753) are told to be use only IHC-P, but I´d like to use them in frozen sections. Is it possible. What´s the difference between antibodies for IHC-F and IHC-P Thank you for the attention

Thank you for your enquiry.

It may indeed be possible to use antibodies in IHC-Fr (frozen sections) when they are already working in IHC-P. This is likely as the fact they work in IHC-P means they have the capacity to recognise the antigen in its native (3 dimensional) form. You may have to try different fixatives to see which one will work. In theory if the antibodies worked with formalin fixed paraffin embedded tissue they are resistant to formalin so a fixation with formaldehyde may work.

The IHC-F application is an old application which we are currently removing, it is the same as IHC-P, I apologise for the confusion.

Thank you for your answer to my question. I have detected the Cytokeratin protein using your ab7753 pan Cytokeratin antibody [C-11]. Next I want to detect the Cytokeratin mRNA in breast cancer. Since your antibody can detect the Cytokeratin 4?5?6?8?10?13?18, so I want to search the common conservative mRNA sequence of the Cytokeratin family. Is it possible? I am looking forward to listening to your comment. Thank you!

It is probably possible. You may need to conduct a search on different databases (GeneCard, Omim, Unigene, Locus) to find out the common conservative mRNA sequence of the Cytokeratin family.

We earlier used the Boehringer Mannheim anti-pan cytokeratin (clone Lu5) for detecting the cytokeratins in the chicken (or other avian species). Now, this mAb is not produced by the BMannheim (as I heard they sold it to an another company). Therefore I am interested if theab7753 does react with avian cytokeratins or not? The PCK 26 (sold by Sigma) did not work well (or at all) in some chicken epithelia, like e.g. the bursal epithelium. Yours sincerely, dr. Attila Magyar

ANSWER:

 

To our knowledge this antibody ahs not been tested on avaian tissue. However, considering the wide species cross-reactivity of this antibody, it may be suitable for chicken as well.
