    The PAP pen is a special marking pen that provides a thin film-like green-tinged hydrophobic barrier when a circle is drawn around a specimen on a slide.

    This water repellent barrier keeps staining reagents localized on the tissue sections, prevents mixing of reagents when differentially staining two sections on the same slide, and allows use of less reagents per section.

    Insoluble in alcohol and acetone, PAP Pen markings can be optionally removed with xylene after the staining procedure is complete.

    PAP Pen is good for approximately 500 uses.

This PAP pen is very easy to use. It dries quickly. It is strong enough to create and maintain a boundary between tissue sections. I usually stain secondary antibody control on the same slide with primary antibody, so I divided the section with the PAP pen. After overnight incubation with antibody, the boundary still functional and there was no mixing between adjacent drop of solutions. Picture was taken after overnight incubation.

PAP pen dries quickly and forms a hydrophobic layer that is not washed off by 0.1% Triton X-100 in PBS. The only concern I have is that the barrier is often difficult to dry after washing, so extra care must be taken to avoid mixing reagents between adjacent sections.

I am happy to confirm that the tip of the Pap pen is about 3 mm thick and tapers off to about 1 mm.

Precise and reliable barrier pen. After application it dries rather quick and after that there's no leakage. Found it very useful for IHC when I had to isolate from each other few tissue samples on the same slide. It resists incubation in buffers with Tween-20. Soluble in Xylene.
